The role of alpha-ketoglutarate and mesaconate in glutamate fermentation by Clostridium tetanomorphum.

چکیده
The products of L-glutamate fermentation by resting or growing cells of Clostridium tetanomorphum are acetate, butyrate, hydrogen, and approximately stoichiometric amounts of carbon dioxide and ammonia (14). Experiments with C14-labeled glutamate have shown that acetate, butyrate, and carbon dioxide originate preferentially from glutamate carbons 1 and 2, 3 and 4, and 5, respectively, and that glutamate carbon atom 4 is the precursor of butyrate carbons 1 and 3 (5, 6). These findings are consistent with a path of glutamate fermentation that involves the formation of 2 different. CZ units, one active in acetate formation and the other a precursor of butyrate, and exclude the operation of a tricarboxylic acid cycle in the oxidative direction. Experiments on the mechanism of this fermentation carried out, with cell-free preparations have furnished evidence for the involvement of both a-ketoglutaric and mesaconic acids.

Stereochemistry of the glutamate mutase reaction.
4-Deuterioglutamic acid was prepared by incubation in DzO of ammonium mesaconate with an extract of Clostri&urn tetanomorphum. In these extracts, methylaspartase catalyzed the formation of threo-3-methyl-L-aspartate-3-D, which was then rearranged to glutamate by the cobamide coenzyme-dependent glutamate mutase. The monodeuteriosuccinate obtained by chloramine-T oxidation of the glutamate showed...

The gene encoding component E, the large subunit, of adenosylcobalamin (coenzyme B12)-dependent glutamate mutase from Clostridium tetanomorphum has been cloned and sequenced. The mutE gene encodes a protein of 485 amino acid residues, with M(r) 53,708. The mutE gene is situated some 1,400 bp downstream of the mutS gene, which encodes the small subunit of glutamate mutase. Between the two is an ...
متن کاملThe metabolism of itaconic acid by liver mitochondria.
The natural occurrence of a great variety of methyl and methylene-substituted dicarboxylic acids has prompted an investigation of the metabolism of some of these compounds by animal tissues. Guinea pig liver mitochondria did not catalyze measurably the oxidation of citraconate, dl-citramalate, P-methyl malate, itamalate, or paraconate. Itaconic acid’ was oxidized as rapidly as most members of t...

متن کاملFormation of n-Butanol from d-Glucose by Strains of the "Clostridium tetanomorphum" Group.
A clostridial strain has been isolated that produced n-butanol, ethanol, butyrate, and acetate as major fermentation products from glucose but no acetone. At a pH of 6.6, n-butanol was formed by this microorganism only during growth. On the basis of its physiological characteristics and DNA-DNA homology data, the strain was assigned to the "Clostridium tetanomorphum" group (S. Nakamura, I. Okad...
